Progesterone and Androstenedione Are Important Follicular Fluid Factors Regulating Porcine Oocyte Maturation Quality

چکیده

Oocytes matured in vitro are useful for assisted human and farm animal reproduction. However, the quality of oocytes is usually lower than that vivo oocytes, possibly due to absence some important signal regulators vitro. In this study, untargeted metabolomics was used detect changes metabolites follicular fluid (FF) during pig oocyte maturation culture medium maturation. Our results showed total metabolite changing profile FF different from medium, but levels 23 differentially expressed (DEMs) changed by following same trend both These may be porcine We found progesterone androstenedione, two factors ovarian steroidogenesis pathway enriched DEMs, were upregulated The these 31 20 fold, respectively, they higher at mature stage. supplementation androstenedione significantly improved rate subsequent embryo developmental competence. finding suggests a metabolic abnormality affects oocytes. This study identified regulate their potential, which will helpful improve

The Effects of Progesterone on Oocyte Maturation and Embryo Development

Oocyte maturation and embryo development are controlled by intra-ovarian factors such as steroid hormones. Progesterone (P4) exists in the follicular fluid that contributes to normal mammalian ovarian function and has several critical functions during embryo development and implantation, including endometrial receptivity, embryonic survival during gestation and transformation of the endometrial...

Hypoxanthine is the principal inhibitor of murine oocyte maturation in a low molecular weight fraction of porcine follicular fluid.

Studies were carried out to identify and quantify the porcine follicular fluid (PFF) component(s) responsible for inhibition of murine oocyte maturation. A low molecular weight fraction of PFF (less than 1000) was prepared by dialysis and used in all experiments. This PFF fraction contained an inhibitor(s) of mouse oocyte maturation that absorbed maximally in the ultraviolet (UV) range at 250-2...

Porcine oocyte maturation in vitro: role of cAMP and oocyte-secreted factors – A practical approach

Polyspermy or the penetration of more than one sperm cell remains a problem during porcine in vitro fertilization (IVF). After in vitro culture of porcine zygotes, only a low percentage of blastocysts develop and their quality is inferior to that of in vivo derived blastocysts. It is unknown whether the cytoplasmic maturation of the oocyte is sufficiently sustained in current in vitro maturatio...

Differences in the metabolomic signatures of porcine follicular fluid collected from environments associated with good and poor oocyte quality.

The microenvironment of the developing follicle is critical to the acquisition of oocyte developmental competence, which is influenced by several factors including follicle size and season. The aim of this study was to characterise the metabolomic signatures of porcine follicular fluid (FF) collected from good and poor follicular environments, using high-resolution proton nuclear magnetic reson...

Energy Metabolism Regulating Mammalian Oocyte Maturation

Oocyte maturation is a final step of gamete development that follows a prolonged period of cell growth within a growing follicle. Unlike oocyte growth that takes several weeks or months, maturation process is a short (hours or days), dynamic process. The period of oocyte maturation varies among species, ranging from 10-13 hours in the mouse (Edwards & Gates, 1959) to 16-24 hours in the cow (Dom...
